These tests have been developed to further our understanding of spermatogenesis and sperm function, and to potentially provide a more accurate infertility diagnosis than by evaluation of standard sperm parameters alone. Conventional sperm parameters such as sperm concentration, motility and morphology exhibit a high degree of biological variability and are only fair measures of fertility potential. Sperm DNA integrity tests have also been developed in the hope that they may predict pregnancy outcome after assisted reproductive technologies (ARTs), as conventional sperm parameters are poor predictors of ART outcomes. Animal models have played a major role in providing strong evidence that sperm DNA fragmentation is highly correlated with male fertility potential. Moreover, experimental studies have demonstrated that sperm DNA damage is associated with poor reproductive outcomes (lower pregnancy rates, chromosomal abnormalities, pregnancy loss, reduced longevity and birth defects) after ARTs. These experiments have raised some concerns regarding the use of DNA damaged sperm in the context of ARTs. However, these observations may or may not translate to equivalent clinical effects because unlike sperm DNA damage in humans, DNA damage in animal models is induced experimentally and is present in all spermatozoa. Several assays have been developed to measure sperm DNA and chromatin damage. The most common are SCSA (sperm chromatin structure assay), COMET (single cell gel electrophoresis) assay, TUNEL (Terminal deoxynucleotidyl transferasemediated dUTP Nick End-Labeling) assay and SCD (sperm chromatin dispersion) assay. It is important to consider that some assays have undergone more rigorous testing than others, that different assays measure different aspects of sperm DNA and chromatin integrity, and that these assays do not selectively differentiate clinically important from insignificant damage. Moreover, sample preparation and handling, and assay conditions (eg accessibility

P-23: Evaluation of Relation between Sperm DNA Fragmentation with IUI Outcomes
Background: Sperm chromatin integrity assessment has been suggested as fertility predictor. The aim of this study was to examine the relationship between the results of sperm chromatin structure assay (SCSA) and the outcome of intrauterine insemination IUI. Materials and Methods: A total of 576 consecutive couples undergoing IUI were included and IUI was performed. SCSA results were expressed a...
متن کاملP-134: In Vitro Assessment of Cysteine Effect on DNA Integrity of Frozen-Thawed Buffalo Spermatozoa
Background: Cysteine has been used as additive in cryopreservation extenders. It contains of thiol group which plays an antioxidant role to eliminate reactive oxygen species (ROS) during freeze -thaw procedure. Excessive ROS production cause DNA damage and subsequently reduce sperm fertility. On the other hand, protamine protein have high levels of cysteine which is critical in packaging and co...
متن کاملP-23: Is There An Association between HOST Grades and Sperm Quality
Background: Intracytoplasmic sperm injection (ICSI) can be considered the solstice in vitro insemination technique because it has powerfully allowed the treatment of male factor infertility, but in this procedure, except visual morphological selection, there is no standardization for sperm selection. Recently, hypo osmotic swelling test (HOST) has been proposed to have the potential to select i...
متن کاملP-77: HOST Patterns and Sperm Quality
Background: Following the expansion of human infertility, different procedures have been utilized for separation of normal sperm during ICSI (Intra cytoplasmic sperm injection). Hypo-osmotic swelling test (HOST) is one of the tests which have been introduced for assessment of integrity of sperm membrane when confronted with hypo osmotic condition. This test may also reflect intracellular organe...
متن کاملP-5: Assessment of Sperm Chromatin Status in Rat Experimental Varicocele
Background: Varicocele is considered as a common cause of male infertility and can affect spermatogenesis. In addition, it also predisposed sperm to chromatin damage and morphological alternation. Other factors, such as environmental and genetic conditions also predispose sperm to these conditions. To answer this question, we aimed to evaluation of DNA fragmentation and protamine deficiency thr...
متن کاملP-19: Effect of Three Sperm Preparation Methods on Sperm DNA Integrity and Oocyte Fertilization Rate
Background: Techniques of sperm preparation require centrifugation to separate spermatozoa from the seminal plasma. The centrifugation can induce damage to the spermatozoa and increase reactive oxygen species(ROS) productionspecially in leukospermic speciemens that are associated with impaired function and fertilizing capacity. However in ICSI, fertilization can occure with damage DNA sperm but...
